    Regioselective reaction: Synthesis, characterization and pharmacological studies of some new Mannich bases derived from 1,2,4-triazoles

    In the present investigation, a series of new 4[(3-substituted-1H-pyrazol-4-yl)methyleneamino]-5-substituted-2-[(4-methylpiperzine-1-yl)methyl]-2H-1,2,4-triazole-3(4H)-thiones (4) were synthesized by the aminomethylation of 4-(3-substituted-1H-pyrazol-3-yl)methyleneamino-5-substituted-4H-1,2,4-triazole-3-thiols (3) with formaldehyde and N-methylpiperzine. These newly synthesized Schiff and Mannich bases were characterized by IR, 1H NMR, mass spectral data and elemental analyses. These compounds were screened for their antibacterial and antifungal activity. Some of the compounds were found to exhibit significant antimicrobial activity. © 2009 Elsevier Masson SAS.     Synthesis and biological evaluation of aminoketones

    A three-component Mannich reaction of different ketones with aromatic aldehydes and different amines in microwave irradiation under solvent free condition afforded corresponding ?-amino carbonyl compounds in excellent yields. This method proved as a novel and improved modification of the reported three-component Mannich reaction in terms of milder reaction conditions, reaction times, clean reaction profiles, very small quantity of catalyst and simple workup procedure. Newly synthesized ?-aminoketones were characterized by spectral studies. Structure of compound 4a was also confirmed by single-crystal X-ray analysis. All the compounds were screened for their antimicrobial activity by MIC method. Few of the molecules were found to be biologically potent. © 2010 Elsevier Masson SAS. All rights reserved.

    Synthesis of some new pyrazolone derivatives as potent antimicrobial agents

    Invasive microbial infections are major problems around the world, especially in immuno compromised patients. The recent expansion of antimicrobial drug research has occurred because there is a critical need for new antimicrobial agents to treat these life threatening invasive infections. In the present study three series of new substituted pyrazolone derivatives (5a-f, 6a-f and 8a,b) were synthesized by the Knoevenagel condensation reaction of pyrazolones (3a,b) with various substituted carbaldehydes (4a-f, 7). These newly synthesized compounds were characterized by IR, NMR, mass spectra and also by C, H, N analyses. New compounds were screened for their antimicrobial studies against S. aureus, B. subtilis, E. coli and P. aeruginosa. The results revealed that compounds 5c and 6c having 2,5-dichlorothiophene substituent showed significant antibacterial activity against all tested microorganisms as compared to the standard drug Ceftriaxone.

    A novel series of homoallylic amines as potential antimicrobials

    An efficient catalytic three-component reaction of aldehydes, amines, and allyltributylstannate has been successfully developed to produce homoallylic amines at 25°C, in excellent yields, in the presence of 1 mol% of trifluoroacetic acid an inexpensive and environmentally friendly catalyst. Newly synthesized compounds were confirmed by spectral studies. Compound 3o was characterized by single crystal X-ray analysis. All the compounds were also screened for their antimicrobial activity. Halogen-substituted compounds namely 3d, 3g, 3n, and 3o have showed excellent antibacterial activity. © Springer Science+Business Media, LLC 2011.

    T3P mediated synthesis of some new quinoline substituted pyrazole derivatives and its antibacterial studies

    A series of new N-(substituted)-5-phenyl-1-(quinolin-2-yl)-1H-pyrazole-4-carboxamide derivatives (5a-h) have been synthesized by condensing ethyl-3-(dimethylamino)-2-[(phenyl)carbonyl]prop-2-enoate 2 with quinoline-2-yl-hydrazine. Compound 3 is hydrolyzed into 4 which is upon further coupled with different aromatic/aliphatic amines using 50% prpopyl phosphonic anhydride (T3P) in ethyl acetate as coupling reagent to afford different quinoline substituted pyrazole carboxamide derivatives. All the newly synthesized compounds were screened for their invitro antibacterial studies against Escherichia coli, Bacillus subtilis and Pseudomonas aeruginosa. The results revealed that compounds 5d, 5f, and 5h showed good antibacterial activity towards all bacterial strains. These newly synthesized compounds were characterized by NMR, mass spectral, IR spectral study and also by C, H, N analyses.

    Design and regioselective synthesis of trifluoromethylquinolone derivatives as potent antimicrobial agents

    Three series of new trifluoromethyl substituted quinolone derivatives were synthesized (4a-f, 6a-f and 8a-f) from corresponding substituted anilines by multi-step reactions. The regioselective alkylation with different alkyl halides were carried out by approaching two different routes to get the final products in good yield. Newly synthesized compounds were characterized by spectral study and also by C, H, N analyses. Three dimensional structure of 2b and 4b were also confirmed by single crystal X-ray studies. The final compounds (4a-f, 6a-f and 8a-f) were screened for their in-vitro antibacterial and antifungal activity by well plate method (zone of inhibition). The results revealed that, compounds 4a, 6b, 6c and 8e showed significant antibacterial activity as compared to the standard drug Ciprofloxacin.     New pyrazole derivatives containing 1,2,4-triazoles and benzoxazoles as potent antimicrobial and analgesic agents

    Azole class of compounds are well known for their excellent therapeutic properties. Present paper describes about the synthesis of three series of new 1,2,4-triazole and benzoxazole derivatives containing substituted pyrazole moiety (11a-d, 12a-d and 13a-d). The newly synthesized compounds were characterized by spectral studies and also by C, H, N analyses. All the synthesized compounds were screened for their analgesic activity by the tail flick method. The antimicrobial activity of the new derivatives was also performed by Minimum Inhibitory Concentration (MIC) by the serial dilution method. The results revealed that the compound 11c having 2,5-dichlorothiophene substituent on pyrazole moiety and a triazole ring showed significant analgesic and antimicrobial activity. © 2012 Elsevier Masson SAS.     Synthesis, and antitubercular and antimicrobial activity of 1?-(4-chlorophenyl)pyrazole containing 3,5-disubstituted pyrazoline derivatives

    A new series of 1?-(4-chlorophenyl)-5-(substituted aryl)-3?-(substituted aryl)-3,4-dihydro-2H,1?H-[3,4?]bipyrazolyl derivatives (6a-e, 8a-e, 10a-e) have been synthesized, characterized and screened for antimicrobial and antitubercular activity. Among the synthesized compounds, the minimum inhibition concentration of 10e was found to be as low as 1.56 ?g ml-1 and that of 10c was 6.25 ?g ml-1 as compared to the standard anti-tb drugs pyrazinamide and streptomycin. © The Royal Society of Chemistry and the Centre National de la Recherche Scientifique 2016.

    Antitubercular and antimicrobial activity of nh4vo3 promoted 1,4-dihydropyridine incorporated 1,3,4-Trisubstituted pyrazole

    Background: A new series of pyrazole containing 1,4-dihydropyridine derivatives 5a-i and 6a-i were synthesized from substituted acetylated aryls and substituted phenylhydrazine by the multistep reaction. Method: The target compounds 1,4-dihydropyridine derivatives were obtained from green synthesis of 1,3-disubstituted phenyl-1H-pyrazole-4-carbaldehydes 4a-i with ethyl acetoacetate and methyl acetoacetate at higher temperatures in the presence of ammonium acetate and the catalytic amount of ammonium metavanadate (NH4VO3). The role of ammonium metavanadate was increases rate of the reaction and obtained high yields. Result: Structures of newly synthesized 1,4-dihydropyridine moiety containing pyrazole derivatives were confirmed by FT-IR, NMR and Mass spectral studies. The structure of compound 5b was confirmed by S-XRD study. Further, these compounds were tested for in-vitro antitubercular and antimicrobial studies. Compounds 5a, 5b, 5i, 6a, 6b, 6g, 6h, and 6i were found to be active against all the bacterial microorganisms. Conclusion: The above mentioned compounds have shown lowest MIC ranging between 3.12-12.5 ?g/ml against Mycobacterium tuberculosis and MIC values ranging between 7.8- 15.6 ?g/ml for Mycobacterium smegmatis, Staphylococcus aureus and Pseudomonas aeruginosa. For the control of life threatening diseases such as tuberculosis, these eight compounds may be strongly promising synthetic compounds. © 2017 Bentham Science Publishers.
