Item Synthesis, characterization and anti-microbial studies of some novel 2,4-disubstituted thiazoles(2010) Vijesh, A.M.; Isloor, A.M.; Prabhu, V.; Ahmad, S.; Malladi, S.In the present study a series of novel 2,4-disubstituted thiazole derivatives containing substituted pyrazole moiety was synthesized by the reaction of 3-Aryl-1H-pyrazole-4-carbaldehyde thiosemicarbazone with 6-Bromo/H-3-(bromoacetyl)-2H-chromen-2-one/phenacyl chloride. Structures of newly synthesized compounds were characterized by spectral studies. New compounds were screened for their antibacterial studies against Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Pseudomonas aeruginosa. The results revealed that compounds 7c, 8e, and 8f having 2,5-dichlorothiophene substituent and 8c, 8d having 2,4-dichlorophenyl substituent showed significant antibacterial activity against all tested microorganisms. © 2010 Elsevier Masson SAS. All rights reserved.Item Synthesis and antimicrobial activity of some new pyrazole containing cyanopyridone derivatives(2012) Malladi, S.; Isloor, A.M.; Peethambar, S.K.; Ganesh, B.M.; Goud, P.S.K.A series of new 4,6-disubstituted-3-cyano-2-pyridone derivatives (4a-o) were synthesized. The structures of all target molecules (4a-o) have been confirmed by various spectral techniques and elemental analyses. The newly synthesized compounds were screened for antibacterial and antifungal activity and most of the compounds showed significant activity comparable with that of the standard drug. The results revealed that 4b, 4c, 4d, 4g, 4m, 4n and 4o showed good antibacterial activity towards all bacterial strains (Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa) when compared to standard drug streptomycin. Amongst all the compounds, 4c showed moderate antifungal activity against Aspergillus flavus. The acute toxicity study has also been carried out for biologically active compounds and the experimental studies revealed that compounds were safe up to 2000 mg/kg and no deaths of animals were recorded.Item Synthesis, characterization and antimicrobial activity of novel ethyl 1-(N-substituted)-5-phenyl-1H-pyrazole-4-carboxylate derivatives(2012) Chandrakantha, B.; Isloor, A.M.; Shetty, P.; Isloor, S.; Malladi, S.; Fun, H.-K.In the present study, a novel series of Pyrazole derivatives (3a-m) were synthesized by condensing ethyl-3-(dimethylamino)-2-(phenylcarbonyl)prop-2- enoate with different aromatic and aliphatic hydrazines. These newly synthesized compounds were characterized by NMR, mass spectral, IR spectral studies as well as by C, H, and N analyses. All the newly synthesized compounds were screened for their antibacterial properties against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Pseudomonas aeruginosa. Among the screened samples, 3c, 3f, 3k, and 3l have showed excellent antibacterial activity against all the tested bacterial strains as compared to the standard drug Ceftriaxone. Few of the compounds were found to be biologically potent. Molecular structure of compound 3i was confirmed by single crystal X-ray analysis. © Springer Science+Business Media, LLC 2011.Item Synthesis and anti-inflammatory evaluation of some new 3,6-disubstituted-1, 2,4-triazolo-[3,4-b]-1,3,4-thiadiazoles bearing pyrazole moiety(2012) Malladi, S.; Isloor, A.M.; Shetty, P.; Fun, H.-K.; Telkar, S.; Mahmood, R.; Isloor, N.In the present study, a new series of 3,6-disubstituted- 1,2,4-triazolo-[3,4-b]-1,3,4-thiadiazoles (4aj) have been synthesized by condensing 3-substituted-4-amino-5- mercapto-1,2,4-triazoles (1a-b) with various 3-substitutedpyrazole- 4-carboxylic acids (3a-e) in the presence ofPOCl3. The structures of newly synthesized compounds were characterized by elemental analysis, IR, 1H NMR, 13CNMR, and mass spectroscopic studies. Structure of the compound 4b was also confirmed by recording the single crystal X-ray structure. All the synthesized compounds were screened for their anti-inflammatory activities by carrageenan induced paw edema method. Anti-inflammatory screening indicated that, compounds 4d, 4e, and 4h were found to be biologically active whereas remaining compounds showed poor antiinflammatory activity. Also molecular docking studies were also performed for compounds which showed good antiinflammatory activity. © Springer Science+Business Media, LLC 2011.Item Synthesis and evaluation of antioxidant, antimicrobial activities of new 2,4-disubstituted thiazoles(2012) Malladi, S.; Isloor, A.M.; Peethambar, S.K.; Shivananda, K.N.A new series of 2,4-disubstituted thiazole derivatives were synthesized by the reaction of various 3-aryl-1H-pyrazole-4- carbaldehyde thiosemicarbazones (2a-e) and phenacyl bromides. The compounds (3a-n) were characterized by IR, NMR, mass spectra and C, H, N analyses. All the synthesized compounds were screened for their antioxidant and antimicrobial activities. Antioxidant studies revealed that, compounds 3b, 3j and 3n showed significant scavenging activity whereas compounds 3e and 3n exhibited significant antimicrobial activity. The acute oral toxicity study for the compound 3e and 3n have revealed good safety profile till the uppermost dose (2000 mg/kg).Item Synthesis, characterization, anticancer, and antioxidant activity of some new thiazolidin-4-ones in MCF-7 cells(2013) Isloor, A.M.; Dhanya, D.; Shetty, P.; Malladi, S.; Pai, K.S.R.; Maliyakkal, N.There are limited studies centring on the potential of thiazolidin-4-ones as anticancer agents. In this study, a new series of 2-(3-substituted-1H- pyrazol-4-yl)-3-(3-substituted-5-sulfanyl-1,2,4-triazol-4-yl)-1, 3-thiazolidin-4-one (4a-o) have been synthesized by cyclo-condensation reaction of 5-substituted-4-[(3-substituted-1H-pyrazol-4-ylmethylidene)amino]-2H-1,2,4- triazole-3-thione (3a-o) and thioglycolic acid. The structures of all the synthesized compounds were confirmed by elemental analysis, spectral techniques like IR, 1H NMR, and mass spectroscopy. Few compounds exhibited dose-dependent cytotoxic effect in MTT assay in human breast cancer (MCF-7) cells. Apoptotic degradation of DNA due to action of potent thiazolidin-4-ones was analysed by agarose gel electrophoresis and visualized by ethidium bromide staining (comet assay). A concentration-dependent increase in tail length and olive tail moment was observed when treated with thiazolidin-4-ones. In vitro antioxidant studies like DPPH and ABTS-free radical scavenging assays-indicated moderate activity of thiazolidin-4-ones. © 2012 Springer Science+Business Media, LLC.Item Synthesis of new 3-aryl-4-(3-aryl-4,5-dihydro-1H-pyrazol-5-yl)-1-phenyl-1H- pyrazole derivatives as potential antimicrobial agents(2013) Malladi, S.; Isloor, A.M.; Peethambar, S.K.; Fun, H.-K.New (E)-1-aryl-3-(3-aryl-1-phenyl-1H-pyrazol-4-yl)prop-2-en-1-ones 6 (pyrazolic chalcones) were synthesized from Claisen-Schmidt reaction of 3-aryl-1-phenylpyrazol-4-carboxaldehydes 4 with several acetophenone derivatives 5. Subsequently, the cyclocondensation reaction of chalcones 6 with phenylhydrazine in acetic acid medium afforded the new 3-aryl-4-(3-aryl-4,5- dihydro-1H-pyrazol-5-yl)-1-phenyl-1H-pyrazoles 7. The synthesized compounds were characterized by spectral studies and evaluated for their in vitro antibacterial activity against three pathogenic bacterial strains, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa, and in vitro antifungal activity against three pathogenic fungal strains Aspergillus flavus, Chrysosporium keratinophilum, and Candida albicans. © 2012 Springer Science+Business Media New York.
